The MDR1 gene defect occurs primarily in dog breeds related to the “Collie” breed and is thought to have its origin in a single animal in the 19th century breeding line. As a result of the defect, the P-glycoprotein is either insufficiently produced or not produced at all, which has considerable effects on the function of the blood-brain barrier, kidneys, intestines, liver and hematopoiesis in the bone marrow. The reason for the lack of P-glycoprotein is the lack of a few base pairs in the dog’s DNA. These animals can show severe neurotoxic reactions when taking various medications.
